Ovalbumin (OVA)-induced asthma model
Ovalbumin (OVA)-induced asthma model
Background
OVA-induced rat asthma model recapitulates many key features of allergic bronchial asthma, such as elevated specific IgE, edema, mucus hypersecretion, early- and late-phase asthmatic responses, increased and sustained eosinophilia and nonspecific AHR, eosinophilic infiltration, increased Th2 cytokines, and decreased interferon-γ (IFN-γ) secretion.
The rat asthma model shares many similarities with human asthma:
(1) Allergic responses are both IgE-mediated;
(2) Biphasic responses consisting of immediate and late-phase reactions readily occur after challenge;
(3) The onset time of the late-phase reaction is similar to that in asthmatic patients.
Materials and methods
SD rats (both male and female, 6–8 weeks old)
Modeling method: Asthma model was established by intraperitoneal injection of OVA + aluminum adjuvant emulsion, followed by nebulization with 5% OVA in an inhalation chamber.
Evaluation indicators: Inflammatory cell count in bronchoalveolar lavage fluid (BALF), ELISA (IL-5 in BALF, serum IgE), HE staining and pathological scoring
