High-fat diet combined with STZ and dorsal skin excision for establishment of type 2 diabetic wound model
High-fat diet combined with STZ and dorsal skin excision for establishment of type 2 diabetic wound model
Background
In diabetic patients, microvascular and peripheral neuropathy can lead to cutaneous nutritional disorders, resulting in skin injury, secondary infection, and other conditions. Once infected, the wound repair capacity is extremely poor, often causing intractable ulcerative wounds.
Chronic skin ulcers are common surgical diseases, among which diabetic skin ulcers represent one type, characterized by repeated episodes and persistent non-healing. In diabetic patients, skin infection can exacerbate diabetic symptoms and is difficult to control. Without timely treatment, the lesion may progress deeply, even leading to amputation.
In this study, a type 2 diabetes mellitus model was first established, followed by creating a dorsal wound to construct a diabetic wound model.
Materials and methods
SD rats (male, 5–6 weeks old)
Modeling method: A type 2 diabetes mellitus model was established by high-fat diet combined with intraperitoneal injection of STZ. On this basis, a wound was created on the back to construct a diabetic wound model.
Positive drug: Jing Wan Hong Ointment
Evaluation indicators: ELISA assay, HE staining, Masson staining, TUNEL staining
