Bovine type Ⅱ collagen-induced rheumatoid arthritis model /CIA model
Bovine type Ⅱ collagen-induced rheumatoid arthritis model /CIA model
Background
As an animal experimental model of rheumatoid arthritis (RA), CIA exhibits multiple peripheral arthritis, local redness and swelling of joints, and severe cases lead to joint deformity. Pathological changes include proliferative synovitis, destruction of articular cartilage, bone erosion, and inflammatory cell infiltration in the joint cavity. High titers of IgG antibodies against endogenous type II collagen can be detected in vivo. These clinical manifestations and laboratory parameters are closely correlated with human RA, making it an ideal model for screening and studying drugs for the treatment of RA.
Materials and methods
DBA/1 mice (male or female, approximately 20 g)
Modeling method: Primary immunization: subcutaneous injection at the base of the tail of an emulsion of bovine type II collagen and complete Freund’s adjuvant. Booster immunization: subcutaneous injection at the base of the tail of an emulsion of bovine type II collagen solution and incomplete Freund’s adjuvant.
Evaluation indicators:arthritis score, HE staining
